چکیده

The kinetics of glucokinase from rat liver were studied over wide ranges of glucose and MgATP2concentrations. The initial rate shows a co-operative dependence on the glucose concentration, with Hill coefficients in the range 1.2-1.5. The degree of glucose co-operativity increases with the MgATP3concentration, but no co-operativity was detected for the dependence of the rate on the MgATP2concentration. The effects observed occur at physiologically reasonable concentrations of glucose and MgATP2and are consistent with the presumed function of glucokinase in maintaining a constant concentration of glucose in the blood.

Solvent isotope effects on the glucokinase reaction

The solvent isotope effects on the reaction catalysed by rat-liver glucokinase have been studied. At low concentrations of glucose and high concentrations of MgATP2there is an inverse solvent isotope effect of 3.5. At high glucose concentrations there is a normal solvent isotope effect of 1.3. In 'HzO there is positive co-operativity with respect to glucose [Storer, A. C. and Cornish-Bowden, A....

Mechanistic origin of the sigmoidal rate behaviour of rat liver hexokinase D ('glucokinase').

Two recent proposals to account for the kinetic co-operativity of hexokinase D ('glucokinase') from rat liver are examined. A model in which the deviations from Michaelis-Menten kinetics result from a random order of binding of the substrates [Pettersson (1986) Biochem. J. 233, 347-350] accounts satisfactorily for the behaviour as a function of glucose concentrations, but it also predicts obser...

Kinetic evidence for a 'mnemonical' mechanism for rat liver glucokinase.

Inhibition studies of glucokinase were carried out with the products of the reaction, glucose 6-phosphate and MgADP-, as well as with ADP3-, Mg2+ and ATP4-. The results of these, together with those of kinetic studies of the uninhibited reaction described previously [Storer & Cornish-Bowden (1976) Biochem. J. 159, 7-14], indicate that the enzyme obeys a 'mnemonical' mechanism. This implies that...

The Kinetics of Coupled Enzyme Reactions

1. A theory is developed to account for the kinetics of coupled-enzyme reactions without assuming that the second reaction follows first-order kinetics. 2. A simple procedure is described for applying the theory to the practical design ofenzyme assays. 3. The validity of the theory is confirmed for the assay of glucokinase with glucose 6-phosphate dehydrogenase as coupling enzyme. 4. The possib...

The purification in high yield and characterization of rat hepatic glucokinase.

A new improved procedure for the purification of rat hepatic glucokinase (ATP-d-glucose 6-phosphotransferase, EC 2.7.1.2) is given. A key step is affinity chromatography on Sepharose-N-(6-aminohexanoyl)-2-amino-2-deoxy-d-glucopyranose. A homogeneous enzyme, specific activity 150 units/mg of protein, is obtained in about 40% yield. The molecular weight of the pure enzyme was determined by severa...
